Use of ROV's in operation of EAN underwater installation in the North Sea. ArchiMer

The purpose of the paper is to present the state of the Art in use of ROV for assistance in offshore operations. This is done through a review of operations and gained experience during East Frigg Field diverless installation in 1988 and 10 years or under water Inspection, Maintenance and Repair (IMR) on all Fields operated by Elf Aquitaine Norge A/S (EAN). Basic requirements to the types of ROVs are given and the cost-efficiency of the operations are discussed. The potential for further development of the ROVs to replace divers for a large part of annual IMR tasks below water is addressed.

The "CRABE" method is a crack detection technique based on the use of low electromagnetic frequencies. This (patented) method relies on a global injection of current within the structure under inspection. The presence of a crack shall cause a disruption in the current flow, and consequently of the magnetic field off the surface in the vicinity of a defect. Development of genomic probes for detection of hepatitis A virus and enteroviruses in shellfish ArchiMer

The development of molecular biology techniques has modified research procedures for certain viruses, particularly hepatitis A virus (HAV) and enteroviruses. Accordingly, we designed cDNA and RNA genomic probes to detect viruses in shellfish in the environment. Better sensitivity was noted with RNA than cDNA probes: only 17/83 shellfish samples were positive with HAV and enterovirus DNA probes, where as 42/83 were positive with HAV and enterovirus RNA probes.

An intercomparison study of the determination of sulfite in tropical shrimps by the West European Fish Technologists' Association (WEFTA) OMA

Eight WEFTA laboratories participated in two series of intercomparison exercises on sulfite determination in tropical shrimps. Samples of tropical shrimps were spiked with sodium metabisulfite (Na<sub>2</sub>S<sub>2</sub>O<sub>5</sub>) and hydroxymethylsulfonate (HMS) at a level of 25-90 mg SO<sub>2</sub>/kg. Most of the laboratories determined the sulfite content with (modified) methods of Monier-Williams or De Vries et al. The overall mean recovery of sulfite was rather low (47-60%), which may be attributed to an irreversible reaction of sulfite with the tropical shrimps. Low cost production and purification of polyclonal antibodies to staphylococcal enterotoxin A Rev. Microbiol.

An immunization scheme for production of antiserum to staphylococcal enterotoxin A (SEA) is proposed. The reference method of Robbins and Bergdoll was modified to reduce the number of doses and the amount of toxin used per animal. The best immunization scheme used injections in days 0, 8, 24, 59, 62 and 67. The amount of toxin at each injection was 5, 6, 20, 50, 100 and 200<FONT FACE="Symbol">m</font>g, respectively. The total amount of toxin was 381<FONT FACE="Symbol">m</font>g, which corresponded to a reduction of 107<FONT FACE="Symbol">m</font>g in the amount of toxin for each animal when compared to the reference method. First record of Marteilia sp in mussels Mytilus galloprovincialis in Croatia ArchiMer

Marteiliosis is a disease of molluscs caused by Marteilia refringens in Europe and M. sydneyi in Australia. During routine examination of cultured mussels Mytilus galloprovinciallis in the northern Adriatic, the occurrence of Marteilia sp. was recorded with a prevalence of 5 %. This parasite was not detected in flat oysters reared in the same area. The affiliation of the detected parasite in M. galloprovinciallis was confirmed by in situ hybridization using a M. refringens probe, specific at the genus level. DNA of these infected mussels originating from the same area will be used to clarify the taxonomic position of this species within the genus Marteilia using a molecular approach.

Garlic viruses often occur in complex infections in nature. In this study, a garlic virus complex, collected in fields in Brazil, was purified. RT-PCR was performed using specific primers designed from the consensus regions of the coat protein genes of Onion yellow dwarf virus, a garlic strain (OYDV-G) and Leek yellow stripe virus (LYSV). cDNA of Garlic common latent virus (GCLV) was synthesized using oligo-dT and random primers. By these procedures individual garlic virus genomes were isolated and sequenced. Little information is available on viral infections that affect bivalve molluscs. Such a lack of data is due ta a certain inadequacy of the diagnosis methods that are employed when massive mortality events occur. Most laboratories involved in mollusc pathology still analyse samples through light microscopy. World-wide, there is thus currently a lack of information concerning the occurrence of bivalve herpesvituses. This is probably due to the lack of suitable diagnostic tools. The basic method for identification and examination of suspect samples is predominantly histopathology. Capture immunoassay for LT detection produced by enterotoxigenic Escherichia coli in bacterial isolates BJM

A capture enzyme-linked immunosorbent assay (ELISA), which detects LT-I toxin produced by enterotoxigenic Escherichia coli strains, has beendeveloped. This capture assay was performed using the IgG enriched fraction of anti-LT-I antiserum and IgG2b anti-LT-I monoclonal antibody and allowed a clear distinction between E. coli LT-I - producing and non-producing strains. The estimated accuracy of the assay is 78% for sensitivity, 94% for specificity and 92% for efficiency. Thus, the capture immunoassayis a sensitive tool for detection of E. coli, which produces heat-labile enterotoxin, and is suitable for use in clinical laboratories and epidemiological surveys in developing world.

Detection of ostreid herpesvirus-1 (OsHV-1) by PCR using a rapid and simple method of DNA extraction from oyster larvae ArchiMer

A DNA extraction procedure was developed for the detection of ostreid herpesvirus-1 (OsHV-1) using the polymerase chain reaction (PCR) in oyster larvae. The DNA extraction procedure developed was tested on 8 larval samples. Abnormal nuclei with characteristic features associated with OsHV-1 infections were only observed in samples in which the viral DNA was detected by PCR. A previously described competitive PCR method was applied to detect inhibition during PCR reactions. The results show that the method can be used on small amounts of oyster larvae (3 mg) for the detection of OsHV-1 DNA by PCR.

A long term study of bonamiosis in Quiberon bay, France ArchiMer

Bonamiosis was first reported in association with mass mortality of flat oysters, Ostrea edulis, in June 1979 in L'Ile Tudy, Brittany. The disease rapidly spread to all of the oyster farming areas in France but also in other European countries. The French flat oyster production, which already suffered from another protozoan disease, marteiliosis, decreased from 5500 mt in 1979 to less than 2000 mt after 1980. In 2001, about 350 French farms sell 1650 mt of flat oysters. This production mainly relies on natural spat collection which specially occurs in Quiberon bay.
